Biochem/physiol Actions:
The ZFN (zinc finger nuclease) provided in this kit targets the SLC19A1 (solute carrier family 19 member 1) gene. At physiological pH, SLC19A1 is involved in the transfer of reduced folates into the cells. It is also involved in the uptake of the antifolate methotrexate. In osteosarcoma and neuroblastoma, decrease in the activity of SLC19A1 leads to methotrexate resistance. In patients with colorectal cancer, high expression of SLC19A1 reduces risk of recurrent disease (disease-free survival). SLC19A1 might be linked with non-syndromic cleft lip and palate and neural tube defects. Additionally, mutations in SLC19A1 gene are associated with risk of ischemic stroke.
General description: CompoZr Knockout ZFNs create targeted double strand breaks at a gene specific target locus. Through the cellular process of Non-Homologous End Joining, this double strand break can result in modification of the DNA sequence and therefore create a functional knockout of the targeted gene. To provide the best chance of creating a functional gene knockout, CompoZr Knockout ZFNs are targeted to the first 2/3 of the coding region for the gene of interest.
