Biochem/physiol Actions:
The ZFN (zinc finger nuclease) provided in this kit targets the SLCO5A1 (solute carrier organic anion transporter family member 5A1) gene. It is expressed in bone tumors, prostate cancer, breast cancer, drug-resistant small cell lung cancer and liver cancer. The SLCO5A1 protein might be involved in cellular responses associated with the reorganization of the cell morphology, for instance differentiation and migration.
General description: CompoZr Knockout ZFNs create targeted double strand breaks at a gene specific target locus. Through the cellular process of Non-Homologous End Joining, this double strand break can result in modification of the DNA sequence and therefore create a functional knockout of the targeted gene. To provide the best chance of creating a functional gene knockout, CompoZr Knockout ZFNs are targeted to the first 2/3 of the coding region for the gene of interest.

Also included in the kit are custom-designed primers, that allow for determination of the rate of mutation and for screening of individual clones that have the desired mutational event, and positive control DNA that has been modified using the CompoZr Knockout ZFN.
