Biochem/physiol Actions:
The zinc finger nuclease (ZFN) provided in this kit targets the gene encoding E-selectin. This protein, along with P-selectin and L-selectin, initiates recruitment of circulating leukocytes from blood to sites of inflammation in the vascular lining through interaction with specific cell surface associated carbohydrate determinants. It is a pro-angiogenic factor. This protein has also been implicated in the pathogenesis of stroke and atherosclerosis.
General description: CompoZr Knockout ZFNs create targeted double strand breaks at a gene specific target locus. Through the cellular process of Non-Homologous End Joining, this double strand break can result in modification of the DNA sequence and therefore create a functional knockout of the targeted gene. To provide the best chance of creating a functional gene knockout, CompoZr Knockout ZFNs are targeted to the first 2/3 of the coding region for the gene of interest.

Also included in the kit are custom-designed primers, that allow for determination of the rate of mutation and for screening of individual clones that have the desired mutational event, and positive control DNA that has been modified using the CompoZr Knockout ZFN.
